FDPS (farnesyl diphosphate synthase) is a crucial enzyme in isoprenoid biosynthesis. The isoprene biosynthetic pathway provides the cells with cholesterol, ubiquinone, and other nonsterol metabolites. FDPS also plays an oncogenic role through GTPase/AKT axis, rendering it an important therapeutic target for cancers. Inhibitors of FDPS are also being used to treat other diseases, such as bone disorders.

Amyloid precursor protein (APP) and its homolog, the APP-like protein 1 (APLP1), are critical players in Alzheimer’s disease (AD). They both exhibit activities in synapse formation and synaptic plasticity. Except for neurons, APP and APLP1 are widely expressed on many kinds of cells. They are also known to participate in the progression, proliferation, and migration of cancers cells.

BACE1 and BACE2 (β-site APP cleaving enzymes 1 and 2) process APP into neurotoxic Aβ peptides. Both molecules are therapeutic targets for AD. BACE2 is also an emerging target for type II diabetes (T2DM) by regulating pancreatic β cell function.

SAP (Serum amyloid P component, PTX2, APCS) is an important component of the humoral arm of innate immunity. It has been strongly associated with many diseases, such as amyloidosis and fibrosis. SAP/APCS is known to bind to the generic structure of amyloid deposits and possibly to protect them against proteolysis. It also binds avidly to amyloid fibrils in AD.
