PROS1 (Protein S) is a protein coding gene located on human chromosome 3q11.1. PROS1 is also known as PSA, PROS, PS21, PS22, PS23, PS24, PS25, THPH5 and THPH6. The human PROS1 gene encodes a 75123 Da protein containing 676 amino acids. The PROS1 protein is broadly expressed in liver, heart and other tissues. Among its related pathways are Gamma carboxylation, hypusine formation and arylsulfatase activation and Cell surface interactions at the vascular wall. PROS1 is related to calcium ion binding and endopeptidase inhibitor activity. SHBG is an important paralog of PROS1 gene. PROS1 is associated with some diseases, including Thrombophilia Due To Protein S Deficiency, Autosomal Dominant and Thrombophilia Due To Protein S Deficiency, Autosomal Recessive.

        Protein S/PROS1 分子背景

        PROS1, also known as protein S, is a vitamin K-dependent plasma protein that functions as a cofactor for the anticoagulant protease, activated protein C (APC) to inhibit blood coagulation. PROS1 has two isoforms: a free, functionally active form and an inactive form complexed with C4b-binding protein. Besides its anticoagulant function, PROS1 also acts as an agonist for the tyrosine kinase receptors Tyro3, Axl, and Mer. The endothelium expresses Tyro3, Axl, and Mer and produces protein S. The interaction of protein S with endothelial cells and particularly its effects on angiogenesis have not yet been analyzed.
