SNAI2 (Snail Family Transcriptional Repressor 2) is a protein coding gene located on human chromosome 8q11.21. SNAI2 is also known as SLUG, WS2D, SLUGH, SLUGH1, and SNAIL2. The human SNAI2 gene encodes a 29986 Da protein containing 268 amino acids. The SNAI2 protein is ubiquitously expressed in endometrium, ovary and other tissues. Among its related pathways are Regulation of Wnt-mediated beta catenin signaling and target gene transcription and Neural Crest Differentiation. SNAI2 is related to sequence-specific DNA binding and proximal promoter DNA-binding transcription repressor activity, RNA polymerase II-specific. SNAI1 is an important paralog of SNAI2 gene. SNAI2 is associated with some diseases, including Waardenburg Syndrome, Type 2D and Piebald Trait.

SNAI2 is a key regulator of the aggressive phenotype observed in endocrine-resistant breast cancer cells, an independent prognostic biomarker in ER+ advanced breast cancer treated with endocrine therapy, and may be a promising therapeutic target in combination with endocrine therapies in ER+ metastatic breast cancer exhibiting high SNAI2 levels. SNAI2 transformed normal fibroblasts to a CAF-like state and boosted their tumor-supporting role in 3D organotypic culture and in OC xenograft model. SNAI2 drove a transcriptional signature in the mesenchymal subtype of OC that contributed to tumor desmoplasia, which fed back to increase SNAI2 expression and sustain fibroblast activation.
