SMAD5 (SMAD Family Member 5) is a protein coding gene located on human chromosome 5q31.1. SMAD5 is also known as DWFC, JV5-1 and MADH5. The human SMAD5 gene encodes a 52258 Da protein containing 465 amino acids. The SMAD5 protein is ubiquitously expressed in endometrium, ovary and other tissues. Among its related pathways are Signaling by GPCR and Signaling by BMP. SMAD5 is related to DNA-binding transcription factor activity and RNA polymerase II core promoter sequence-specific DNA binding. SMAD1 is an important paralog of SMAD5 gene. SMAD5 is associated with some diseases, including Fibrodysplasia Ossificans Progressiva and Deafness, Autosomal Dominant 52.

        SMAD5 is a member of the SMAD family. Members of this family mediate signal transduction by the TGF-beta/activin/BMP-2/4 cytokine superfamily from receptor Ser/Thr protein kinases at the cell surface to the nucleus. SMAD5 is involved in the TGF-beta signaling pathway that results in an inhibition of the proliferation of hematopoietic progenitor cells. It is also involved in cell signalling and modulates signals of bone morphogenetic proteins (BMP's). The binding of ligands causes the oligomerization and phosphorylation of the SMAD5 protein. SMAD5 is a receptor regulated SMAD (R-SMAD) and is activated by bone morphogenetic protein type 1 receptor kinase.
