ZIC3 (Zic Family Member 3) is a protein coding gene located on human chromosome Xq26.3. ZIC3 is also known as HTX, HTX1, ZNF203 and VACTERLX. The human ZIC3 gene encodes a 50569 Da protein containing 467 amino acids. Among its related pathways are Transcriptional regulation of pluripotent stem cells and Mesodermal Commitment Pathway. ZIC3 is related to DNA-binding transcription factor activity and RNA polymerase II proximal promoter sequence-specific DNA binding. ZIC2 is an important paralog of ZIC3 gene. ZIC3 is associated with some diseases, including Heterotaxy, Visceral, 1, X-Linked and Vacterl Association, X-Linked, With Or Without Hydrocephalus.

    ZIC3, or zic family member 3, is a molecule that regulates early embryonic patterning in vertebrates. ZIC3 has putative roles in a number of developmental signalling pathways that have distinct roles in establishing the left-right axis. In addition, ZIC3 is also considered as target of MiR-564, a tumor suppressor in human lung cancer. Variants in the ZIC3 gene are rare, but have demonstrated their profound clinical significance in X-linked heterotaxy, affecting in particular male patients with abnormal arrangement of thoracic and visceral organs.
