The plasmid was transfected into 293E adherent cells with Sinofection reagent (Cat# STF01). After 48 h, Immunofluorescence staining of cells. Cells were fixed with 4% PFA, permeabilzed with 0.3% Triton X-100 in PBS, blocked with 10% serum, and incubated with Mouse anti-Flag Tag monoclonal antibody (CST#8146S) at 37℃ 1 hour. Then cells were stained with Goat Anti-mouse IgG secondary antibody. The fluorescent signal is detected by fluorescence microscope. Each expression experiment has negative control.
ZDHHC12 (Zinc Finger DHHC-Type Palmitoyltransferase 12) is a Protein Coding gene. The ZDHHC12 gene, located on 9q34.11, is conserved in chimpanzee, dog, cow, mouse, rat, chicken, zebrafish, and frog. ZDHHC12 belongs to the DHHC palmitoyltransferase family. 3 alternatively spliced human isoforms have been reported. ZDHHC12 is widely expressed in the colon, stomach, and other tissues. ZDHHC12-mediated S-palmitoylation promotes the cell membrane localization of claudin-3 (CLDN3) and maintains its stability in ovarian cancer cells, contributing to the oncoprotein function of CLDN3. Targeting ZDHHC12-mediated CLDN3 S-palmitoylation might be a potential strategy for ovarian cancer therapy. 225 organisms have orthologs with the human gene ZDHHC12.
