Certain subgroup C strains of herpesvirus saimiri (HVS), a T-cell tumor virus of New World monkeys, transform human T cells into stable growth in culture. The transformed T cells retain essential functions with few exceptions, such as high gamma-interferon (IFNG; 147570) production levels. By subtractive hybridization of HVS-transformed and nontransformed T-cell lines, Knappe et al. (2000) obtained a cDNA encoding AK155. The deduced 171-amino acid protein, which is 25% identical to interleukin-10 (IL10; 124092), contains a signal sequence, 6 helices, and 4 highly conserved cysteines. Northern blot analysis revealed expression of a 1.3-kb transcript in HVS-transformed T cells but not other T-cell lines or primary stimulated T cells. RT-PCR and Southern hybridization analyses detected AK155 transcripts in a series of leukemia T-cell lines but not in B-cell lines. SDS-PAGE and Western blot analyses of recombinant and endogenous AK155 showed a 19-kD secreted protein that could dimerize to a 36-kD protein.
